From the cave, your enemies are going to attack once you click on the play button, which is going to start off the wave.

What you have to do is place defensive towers along the way, before starting the wave, which is going to attack the enemies automatically when insight. You can place only a few, but after defeating waves and getting money in return, you can use it to buy more weapons, as well as towers.
